If I'm going to put Amsoil synthetic gear lube in the rear diff of my truck (3.73LS), do I have to use friction modifier?

Yes since you have the limited slip rear end. They still recommend using the additive even though you are going with the syn.

Oh you can look this up on Amsoils website and go to the product application guide to get the information on which oil is recommended for which rear end you have.
